In Vitro Assessement of Synergistic Anti-Tumor Effects of Resveratrol and Fisetin In Hepatocellular Carcinoma

Hepatocellular carcinoma (HCC) remains among the most diagnosed types of cancer worldwide, representing approximately 75-80% of total primary liver malignancies and one of the main causes of mortality cancer-related on a global scale. The unsatisfactory response to actual treatment options for HCC, in addition to the inherent resistance, toxicity, and low selectivity of it, redirected the focus on naturally-derived therapies.Resveratrol (RES) and fisetin (FIS) exhibited well-documented benefits in HCC by targeting and modulating the critical signaling pathways involved in carcinogenesis and inhibiting the angiogenesis processes, along with exerting anti-inflammatory, antioxidant, and pro-apoptotic effects. In this context, the present study aims to offer an in vitro evaluation of RES and FIS and the combination of both as a potential alternative approach in HCC therapy.The study was performed on Hep G2 (hepatocellular carcionoma cell line) and HepaRG (normal hepatic cell line) as in vitro models. The cell viability determination was performed applying the MTT technique, the cytotoxicity potential was evaluated by the LDH method, the cell morphology and confluence were analyzed using bright-field microscopy, the nuclear morphology was observed by Hoechst 33342 staining, and the caspase activity was determined in order to evaluate the apoptosis induction. After 24 h of treatment with RES, FIS (20, 40, 60, 80, 100 µM), and the combinations of both, a cell confluence reduction and an increasing cytotoxicity were observed into a dose-dependent manner on Hep G2, being the most significant in the combined treatment. The cell morphology and the aspect of nuclei modifications, as well as the elevated level of caspase activity, were more pronounced in the combined treatment. On HepaRG, RES, FIS and the combinations of both, demonstrated a lower cytotoxicity in comparison with Hep G2.The results of this study highlighted the effect of RES and FIS in cell proliferation regression and aptopotosis initialization, underscoring the promising potential of combined treatment as an alternative therapy in HCC.Key words: Cancer, Resveratrol, fisetin, cytotoxic, pro-apoptotic.
